AMT's MAA for LPLD gene therapy product Glybera on schedule

Amsterdam Molecular Therapeutics, a leader in the field of human gene therapy, today announced that the Marketing Authorisation Application (MAA) for Glybera(R) remains on schedule following meetings with the European Medicine Agency (EMA) concerning the Day 120 List of Questions. The company is confident that Glybera(R), a gene therapy product for lipoprotein lipase deficiency (LPLD), remains on track for a regulatory decision by mid-2011.

Research confirms genetic link to advanced fatty liver disease

Researchers at Massachusetts General Hospital found that patients with nonalcoholic fatty liver disease (NAFLD) who carry an allele of the PNPLA3 gene have an increased risk of developing advanced disease, including nonalcoholic steatohepatitis (NASH), fibrosis, and cirrhosis. A second study supported by the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) validates these findings and further concludes that in pediatric patients, the same allele is associated with earlier disease presentation. Both studies are available in the September issue of Hepatology, a journal published by Wiley-Blackwell on behalf of the American Association for the Study of Liver Diseases (AASLD).

Alnylam publishes research findings of Sort1 gene role in cardiovascular disease development

Alnylam Pharmaceuticals, Inc. a leading RNAi therapeutics company, announced today the publication of new research findings in the journal Nature describing the discovery and validation of the role of the gene Sort1 in the development of cardiovascular disease, including myocardial infarction (MI). This work was done with collaborators at University of Pennsylvania (UPenn) School of Medicine, Massachusetts General Hospital (MGH), and the Broad Institute. The collaborative effort combined genome-wide association studies (GWAS) and RNAi technology to identify and validate novel genes as targets for new therapies for heart disease.

SORT1 pathway holds promise as new target for therapeutic intervention for LDL cholesterol reduction

The true power of genomic research lies in its ability to help scientists understand biological processes, particularly those that - when altered - can lead to disease. This power is demonstrated dramatically in a pair of papers published today in the journal Nature. In the first, a global team of researchers describes 95 different variations across the genome that contribute in different degrees to alterations in blood cholesterol and triglyceride levels in multiple human populations. In the second report, close examination of just one of these common variants not only reveals the involvement of an unexpected genetic pathway in lipid metabolism but also provides a blueprint for using genomic findings to unravel biological connections between lipid levels and coronary heart disease.

Scientists identify 95 gene loci that influence lipoprotein metabolism in humans

In a large-scale international study in which Helmholtz Zentrum München participated, scientists have identified 95 gene loci which influence lipoprotein metabolism in humans. Viewed from a long-term perspective, these findings open up new lines of research in the prevention and treatment of coronary artery disease. It is the first genome-wide association study of this kind to be published in the prestigious journal Nature.

Contrave added to diet and exercise regimen can induce significant weight loss

Orexigen Therapeutics, Inc. announced the online publication of results from its COR-BMOD trial of Contrave (naltrexone SR/bupropion SR) in the journal Obesity. The COR-BMOD trial evaluated intensive behavior modification plus Contrave32 (32mg naltrexone sustained release (SR)/360mg bupropion SR) compared to BMOD plus placebo, in 793 overweight or obese patients.

Scientists unveil new biomarkers for stroke, coronary heart disease

Analysis of blood protein data from the Women's Health Initiative cohorts has revealed new biomarkers for stroke and coronary heart disease (CHD). Research published in BioMed Central's open access journal Genome Medicine found that beta-2 microglobulin (B2M) levels were significantly elevated in postmenopausal women with CHD, and insulin-like growth factor binding protein 4 (IGFBP4) was strongly associated with stroke.

ESC concerned over interpretation of JUPITER sub-analysis trial results about HDL cholesterol

The European Society of Cardiology (ESC) is concerned that interpretations of a paper about cholesterol, published in the Lancet, could act to deter ongoing research efforts into developing new therapeutic strategies to increase high density lipoprotein (HDL) cholesterol. Caution, the ESC experts advise, should be displayed in the interpretation of the results.

Cerenis Therapeutics closes first round of series C, raises €40 million

Cerenis Therapeutics SA (Cerenis), a biopharmaceutical company specializing in the research and development of novel therapies based on the metabolism of HDL (the "good" cholesterol) to treat cardiovascular and other metabolic diseases, announced today it has raised €40 million in the first close of its series C financing. In this round €20 million came from the Fund for Strategic Investment (FSI) with an additional €20 million coming from the existing investors: Sofinnova Partners, HealthCap, Alta Partners and TVM Capital, EDF Ventures, OrbiMed and DAIWA Corporate Investment.

Study suggests chronic kidney disease affects persons of African, Hispanic heritage at higher rate

Chronic kidney disease affects millions in North America, with persons of African heritage being at a four-fold higher risk and those of Hispanic heritage having a two-fold higher risk compared to the rest of the population. An international study carried out by Dr. Karl Skorecki, from the Technion-Israel Institute of Technology and Rambam Medical Center in Haifa, and his team points to the APOL1 gene as involved in the increased risk of kidney disease in this high-risk population. The results are to be published online in Springer's journal Human Genetics.

New findings reported in the July issue of Cell Metabolism, a Cell Press publication, are offering new leads as to why some people might suffer from high levels of triglycerides.
