New data on impact of spike mutations and B117 UK variant on neutralization by mAbs and sera

A recent paper by British and Dutch scientists, currently available on the bioRxiv* preprint server, shows that the neutralizing activity of monoclonal antibodies can be dramatically reduced by mutations of the SARS-CoV-2 spike glycoprotein – unlike polyclonal antibodies generated after natural infection that remain active regardless of any introduced amino acid changes.

A recent study has found that extracellular vimentin can bind to SARS-CoV-2 spike protein in vitro, and antibodies against vimentin can decrease virus infectivity by up to 80%. This suggests vimentin could aid the virus’s binding to host cells and help in infection.

Neutralization antibodies affected by only a few mutations to the SARS-CoV-2 receptor-binding domain

In a recent bioRxiv* preprint paper, US researchers mapped in detail how mutations to the receptor-binding domain (RBD) of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) affected antibody binding in convalescent human serum – and revealed that it is influenced by mutations at only a handful of dominant epitopes.

3D imaging of SARS-CoV-2 infection in ferrets using light sheet microscopy

Researchers from the Friedrich-Loeffler-Institut, Federal Research Institute for Animal Health demonstrate the ability to detect severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection in 3D tissue samples in ferrets. This will allow the detection of infection patterns in tissues and their interrelations.

A map of SARS-CoV-2 escape mutations enables the development of therapeutic antibody cocktails

A recent tour-de-force study by the US researchers describes a comprehensive approach to completely map mutations to the SARS-CoV-2 receptor-binding domain (RBD) that escape antibody binding – enabling, in turn, rational design of antibody therapeutics and appraisal of the antigenic consequences of viral evolution.
