One-Step Concentration and Viability

The Cellometer Auto T4 cell counter concurrently calculates the concentration and % viability of cells for Trypan blue-stained cultured cells.

Being a dye exclusion technique, Trypan blue viability detects dead cells by using membrane integrity. The dye cannot enter healthy cells, and therefore they remain unstained. A compromised cell membrane in dead cells is permeable to the Trypan blue dye. The blue-stained dead cells appear as dark cells in the Cellometer software with bright-field imaging.

Within 10 seconds, the Auto T4 instrument reports:

NCI-60 is a class of 59 human cancer cell lines, which were originally 60 cell lines and created by the National Cancer Institute for screening purposes. The cell lines, extracted from different tissues (such as lung, brain, bone marrow, ovary, kidney, breast, skin, prostate, and colon), are widely used for analyzing the development of tumors and for screening experimental cancer drug compounds.

Clumpy Cells

The MCF-7 breast cancer cell line can be quite clumpy. The Cellometer pattern-recognition software detects and counts each cell present inside these cell clumps for precise analysis, as illustrated below.

Cell Size Analysis and Size-Based Counting

The Cellometer Auto T4 cell counter automatically creates a cell size histogram depending on the diameter of cells.

Since the Cellometer Auto T4 cell counter produces measurements of individual cell size, numerous samples can be superimposed on a single histogram. This helps to analyze the changes that occur in the diameter of the cells over time.

Multiple Cell Images for Data Verification

Each cell is different from the other. With the Cellometer Auto T4 cell counter, users can instantly view cell morphology on-screen.

A couple of images, each with four fields of view are captured for each cell count. This corresponds to the area of four quadrants of a hemocytometer. Furthermore, counted cells are shown on-screen to further verify if cells present in the sample are being imaged and studied correctly.

The % coefficient of variation (CV), such as sample preparation, of the Cellometer Auto T4 cell counter was well within 5% for a majority of cell lines tested. Counting Chambers — No Washing or Contamination

The Cellometer Disposable Counting Chambers feature two separately enclosed chambers with an accurately regulated height. Using a normal single-channel pipette, a 20 µL cell suspension is placed into the chamber. This chamber is then inserted into the Cellometer Auto T4 cell counter and the cells are finally imaged.
