The Cellometer Auto T4 bright-field cell counter available from Nexcelom Bioscience is an automated instrument that enables Trypan blue viability for cell lines.

Cell Counting Made Simple

Cellometer Auto T4

Simple, Automated Cell Counting in Less than 10 Seconds

The Cellometer Auto T4 cell counter uses bright-field imaging and pattern-recognition software to rapidly and precisely detect and count individual cells. Count, diameter, concentration, and % viability of cells are calculated and reported automatically.

Load sample, view image, count cells, and obtain results in less than 10 seconds.

The Cellometer Auto T4 cell counter provides the following features:

  • Potential to record various fields of view for highly precise information
  • On-screen view of individual cell level data
  • Integrated cell type library
  • Options for IQ and OQ validation and GLP/GMP software
  • Helps count difficult cells (irregular-shaped and clumpy)

One-Step Concentration and Viability

The Cellometer Auto T4 cell counter concurrently calculates the concentration and % viability of cells for Trypan blue-stained cultured cells.

Cellometer Auto T4

Being a dye exclusion technique, Trypan blue viability detects dead cells by using membrane integrity. The dye cannot enter healthy cells, and therefore they remain unstained. A compromised cell membrane in dead cells is permeable to the Trypan blue dye. The blue-stained dead cells appear as dark cells in the Cellometer software with bright-field imaging.

Within 10 seconds, the Auto T4 instrument reports:

  • Live and total concentration of cells
  • Dead, live, and total count of cells
  • % viability
  • Mean diameter of cells

Analyze More than 99% of Cultured Mammalian Cells, including NCI-60 and clumpy MCF-7 cells

NCI-60 is a class of 59 human cancer cell lines, which were originally 60 cell lines and created by the National Cancer Institute for screening purposes. The cell lines, extracted from different tissues (such as lung, brain, bone marrow, ovary, kidney, breast, skin, prostate, and colon), are widely used for analyzing the development of tumors and for screening experimental cancer drug compounds.

Irregular-Shaped Cells

Users can adjust the Cellometer cell roundness setting to detect and count irregular-shaped cells, like activated T-cells and RD cells.

Cellometer Auto T4

Clumpy Cells

The MCF-7 breast cancer cell line can be quite clumpy. The Cellometer pattern-recognition software detects and counts each cell present inside these cell clumps for precise analysis, as illustrated below.

Cellometer Auto T4

  • Of the NCI-60 cell lines, 57% cells are clumpy, display huge differences in cell size or shape, or contain debris
  • The entire 59 NCI-60 cell lines have been effectively verified on the Cellometer Auto T4 cell counter

Cell Size Analysis and Size-Based Counting

The Cellometer Auto T4 cell counter automatically creates a cell size histogram depending on the diameter of cells.

Since the Cellometer Auto T4 cell counter produces measurements of individual cell size, numerous samples can be superimposed on a single histogram. This helps to analyze the changes that occur in the diameter of the cells over time.

Cellometer Auto T4

In experiments where SF21 or SF9 cells are used, saved images can be observed and re-counted to examine the morphological changes over time.

Cellometer Auto T4

Settings for the maximum and minimum cell diameter can be improved to count specific cells present in a sample. This example shows the counting of mature dendritic cells grown from PBMCs based on the diameter of cells.

Multiple Cell Images for Data Verification

Each cell is different from the other. With the Cellometer Auto T4 cell counter, users can instantly view cell morphology on-screen.

A couple of images, each with four fields of view are captured for each cell count. This corresponds to the area of four quadrants of a hemocytometer. Furthermore, counted cells are shown on-screen to further verify if cells present in the sample are being imaged and studied correctly.

Users can confirm whether:

  • Debris has been eliminated
  • Cells have been counted properly depending on their shape and size
  • Cells within clumps have been counted individually
  • Cell images have been archived and exported for use in presentations and publications
  • Saved images can be again counted using user-defined or default analysis settings

Exclusion of Debris

The Cellometer Auto T4 cell counter detects cells based on morphology, brightness, and size, and therefore, cellular debris is effortlessly and precisely eliminated from counting results.

  • Cell size parameters can be altered to improve the removal of debris from results and also improve the precision of counting for cells of different sizes
  • The counted image can be visualized to confirm the removal of debris from results

10x Faster and More Precise than Manual Counting

It takes around 5 minutes to count 1 x 106 cells with a manual hemacytometer. Sometimes, it takes twice as long to count both dead and live cells. The Cellometer Auto T4 cell counter determines cell count and concentration for dead cells and live cells and also calculates % viability in only 10 seconds.

Cellometer Auto T4

Cellometer Precision

The % coefficient of variation (CV), such as sample preparation, of the Cellometer Auto T4 cell counter was well within 5% for a majority of cell lines tested. In the case of manual counts, the CVs spanned from 7% to 20%. Inter-operator variability and subjectivity from cell counts are removed by automated cell counters to produce more accurate data.

Data Accuracy and Consistency can be Improved

  • Recording and calculation errors are eliminated
  • Judgment errors are eliminated
  • Wash steps are eliminated
  • Counting time is reduced, enabling users to run more experiments

Counting Chambers — No Washing or Contamination

The Cellometer Disposable Counting Chambers feature two separately enclosed chambers with an accurately regulated height. Using a normal single-channel pipette, a 20 µL cell suspension is placed into the chamber. This chamber is then inserted into the Cellometer Auto T4 cell counter and the cells are finally imaged. This technique of sample loading and analysis is easy and perfect for delicate cells.

Cellometer Auto T4

The disposable Cellometer cell counting chambers provide many major benefits:

  • No danger of cross-contamination
  • Elimination of washing step saves time
  • Decreased biohazard risk to users
  • The most economical automated counting consumables
  • Large-depth chambers for massive cells
  • A controlled sample volume can be achieved
