General Notes

The ab135372 is recommended as an alternative by Abcam.

This antibody is ideal for staining neoplastic and normal T cells in formalin-fixed, paraffin-embedded tissues. Abcam-recommended secondaries are Goat Anti-Rabbit Alexa Fluor® 488 (ab150077) and Goat Anti-Rabbit HRP (ab205718).

The Abpromise guarantee provided by Abcam covers the usage of ab16669 in the tested applications given below. Recommended starting dilutions are given below; the end-user must determine optimal dilutions/concentrations.

Application Abreviews Notes
Flow Cyt ☆☆☆☆ 1/1000.
The ab172730 Rabbit monoclonal IgG is appropriate to be used as an isotype control with this antibody.
Abcam recommends using Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody.
IHC-Fr ☆☆☆☆☆ Should be used at an assay dependent concentration
IHC-P ☆☆☆☆ 1/150.
Before starting with IHC staining protocol, heat-mediated antigen retrieval should be carried out with citrate buffer pH 6.
The tissue section should be boiled in 10 mM citrate buffer, pH 6.0 for 10 minutes. Then it should be cooled at room temperature for 20 minutes.
WB ☆☆☆☆☆ 1/200.
Estimated molecular weight: 23 kDa.
IHC-FoFr ☆☆☆☆☆ Should be used at an assay dependent concentration

Signal transduction is mediated by the CD3 complex.

Involvement in Disease

Severe combined immunodeficiency autosomal recessive T-cell-negative/B-cell-positive/NK-cell-positive (T(-)/B(+)/NK(+) SCID) [MIM:608971] is caused due to defects in CD3D.

One form of severe combined immunodeficiency (SCID) is a clinically and genetically heterogeneous group of rare congenital disorders marked by the destruction of both cell-mediated and humoral immunity, leukopenia, and low or no antibody levels.

Children with this disorder experience frequent and persistent infections by opportunistic organisms. The absence of T-cell-mediated cellular immunity because of a defect in T-cell development is the common characteristic of all forms of SCID.
