Before starting with IHC staining protocol, heat-mediated antigen retrieval should be carried out with citrate buffer pH 6.

Severe combined immunodeficiency autosomal recessive T-cell-negative/B-cell-positive/NK-cell-positive (T(-)/B(+)/NK(+) SCID) [MIM:608971] is caused due to defects in CD3D.

One form of severe combined immunodeficiency (SCID) is a clinically and genetically heterogeneous group of rare congenital disorders marked by the destruction of both cell-mediated and humoral immunity, leukopenia, and low or no antibody levels.

Children with this disorder experience frequent and persistent infections by opportunistic organisms. The absence of T-cell-mediated cellular immunity because of a defect in T-cell development is the common characteristic of all forms of SCID.
