Phosphoenolpyruvate Carboxykinase (PEPCK EC 4.1.1.32) is an enzyme belonging to the lyase group. In the presence of GTP, it catalyzes the reversible conversion of oxaloacetate (OAA) into GDP, phosphoenolpyruvate (PEP), and CO2. There are two isoforms of PEPCK found in humans: mitochondrial form (PEPCK-M) and cytosolic form (PEPCK-C, also known as PCK1). PEPCK-C is a rate-regulating step in gluconeogenesis. New studies found irregular concentrations of PEPCK in diabetic mice. Thus, accurate measurement of Phosphoenolpyruvate Carboxykinase activity is advantageous for both therapeutic and mechanistic studies.

The Phosphoenolpyruvate Carboxykinase (PEPCK) Activity Assay kit from BioVision provides a fast and easy way of measuring PEPCK activity in a range of samples. In this assay, Phosphoenolpyruvate Carboxylase is coupled with a set of enzymes that change PEP and carbonate into a series of intermediates and hydrogen peroxide, which in turn, reacts with a probe and converted producing a colorimetric signal (OD: 570 nm). The color intensity is directly proportional to the quantity of active Phosphoenolpyruvate Carboxylase found in the samples. The assay is sensitive, simple, high-throughput adaptable and has the capacity to detect less than 10 µU of Phosphoenolpyruvate Carboxykinase activity per sample.
