Automation of DNA library construction using Beckman Coulter’s Biomek FXP workstation

The manual preparation of numerous sequencing libraries can be a labor-intensive process that occupies a significant amount of researchers’ time. Manual processes are also associated with an increased risk of inconsistency and human error. In order to benefit from Illumina’s powerful next generation sequencing (NGS) technology, efficient and reliable automation solutions are essential for DNA library construction.

The Biomek® FXP Dual-Arm Multi-96 and Span-8 Workstation (BFXP) is a single, compact system that combines every aspect of liquid handling required to automate NGS sample preparation. The system provides high-quality template libraries at the throughput needed to benefit from the high capacity of NGS, while offering the flexibility needed to meet the operator’s changing requirements.  

Scientists from Beckman Coulter have recently described the automation of Illumina’s TruSeq® Nano DNA kit on the BFXP for the interrogation of sample inputs as low as 100 ng. Up to 96 samples can be run simultaneously and the system set-up requires just a few clicks of the mouse and one-time pipetting of kit components into reservoirs. The ease and speed of reaction set-up is improved by the software and pipetting tools.

The BFXP user interface provides the user with various options for selecting the set-up of library construction including which steps are performed, how many samples are prepared, how incubations are handled and which adapter labware is used with which samples. A reagent calculator is also available which can tell the user which reagents are needed, how they should be prepared and what the required reagent volume is.

Using the TruSeq Nano DNA kit on the BFXP, up to 96 high quality, sequence-ready libraries can be constructed from low input samples in just over 8 hours. Consistency between the libraries produced is high in terms of percent alignments to the reference genome and quality scores, the percent PCR duplication is low and the median fragment length between replicates is also highly reproducible.

This automation solution standardizes the NGS sample preparation process, reducing the risk of human error, lowering reagent cost and freeing up valuable time for researchers to walk away from the system and complete other tasks.
