Researchers unveil new SARS-CoV-2 inhibitor that targets viral immune evasion

In a recent quest for an effective antiviral compound, UK researchers have purified a specific enzyme of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) known as nsp15 and optimized fluorescent biochemical endoribonuclease assays to screen a custom chemical library with over 5,000 commercial compounds. The study is currently available on the bioRxiv* preprint server.

A new Morbidity and Mortality Weekly Report published by the U.S. Centers for Disease Control and Prevention (CDC) states that mRNA COVID-19 vaccines are highly effective in preventing SARS-CoV-2 infection in the real world. The report is based on a CDC study where it routinely tested prospective cohorts of health care professionals, first responders, and essential and frontline workers for SARS-CoV-2 infections.

Research led by Michael J. Hannon from the University of Birmingham, UK, finds metallo-supramolecular helicates (cylinders) binding to untranslated regions of SARS-CoV-2 suppresses viral replication. The untranslated regions (UTR) of the viral genome could serve as drug targets for antiviral treatment.

Researchers offer large-scale screening of potential broad-spectrum SARS-CoV-2 antivirals

A new study describes the identification of new compounds from a library, with inhibitory activity against several coronaviruses. This study shows the utility of this method, besides identifying compounds that not only inhibit viral shedding in SARS-CoV-2 cultures, but have an immunomodulatory effect that may be useful in mitigating the damaging cytokine storm typical of severe or critical COVID-19.

Researchers examine circRNAs encoded by MERS-CoV, SARS-CoV-1 and SARS-CoV-2

New research published in Briefings in Bioinformaticsm has identified thousands of circRNAs in Betacoronaviruses, including MERS-CoV, SARS-CoV-1, and SARS-CoV-2, from the RNA-sequencing data of viral infections. These coronaviruses have a similar genomic structure with a diverse circRNA coding ability. Researchers have also systematically characterized circRNAs in coronaviruses, with regard to their size, sequence feature, expressions, genome, location preference, and functions. This opens the door to further explore and understand the mode of action of individual circRNAs in coronaviruses.

Research finds polymerase of SARS-CoV-2 can adopt an alternative structure

New research by German scientists has uncovered another form of coronavirus polymerase. The current polymerase thought to be used for the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a viral RNA-dependent RNA polymerase (RdRp) for replication and transcription. It contains a monomeric enzyme with the catalytic subunit nsp12, two copies of subunit nsp8, and one copy of subunit nsp7.
