Capabilities
One to eight samples can be placed into the Unas, secured in the holder with a twist, and prepared for use. Each Una can accommodate sample volumes ranging from 0.5 to 8 mL. They are constructed with regenerated cellulose (10, 30, and 100 kDa MWCOs) and polyethersulfone (30 and 100 kDa MWCOs) to ensure that the samples remain intact, prevent absorption, and enable efficient liquid passage with a recovery rate of ≥96%.

The secret sauce
Unagi has perfected the UF/DF buffer exchange. It begins by assessing the initial sample volumes using an ultrasonic sensor to understand the starting point. It then applies pressure to each Una while delicately mixing the samples to initiate the flow. It then reevaluates the volume and replenishes each Una with the new buffer. This iterative process continues until Unagi achieves the desired exchange percentage and volume.

Put in order
Unagi offers a user-friendly experience, making it accessible to everyone. The desired experiment type, the molecule being exchanged, the initial volume, and the target buffer can be selected. The final step involves entering the desired percent exchange and, if necessary, the final volume if the sample is being concentrated.

Get the fill
Trays can be loaded with samples at the front. Falcon tubes can be filled with the required buffers and placed on the left side. Tip racks can be easily placed on the right, and Unagi is primed for action.

Let it roll
The process can be initiated by pressing the start button and allowing Unagi to take care of the rest.
Users can monitor Unagi’s progress at their convenience, keeping count of the completed exchange cycles, offering details on the ongoing filtration cycle, and presenting the percentage of exchange achieved for each sample.
Users can also pause the run to retrieve samples that have been completed ahead of the others or let it continue. In any case, Unagi will notify users when the process is complete.

Take it down
Specific samples may be excessively diluted and require some attention. Unagi can concentrate these samples to a more usable range. Opt for the “Reduce Sample Volume” application, and Unagi will efficiently concentrate as many as eight dilute samples, reducing them from 48 mL to 8 mL in a single operation. Following this, users can proceed with a buffer exchange run or further concentration.

Get the check
Unagi meticulously records and logs each action it takes throughout the run, documenting every event. It generates a comprehensive report containing final volumes and concentrations, flow rates for every cycle, and the ultimate percentage of exchange achieved for each sample. These reports can be conveniently integrated into a laboratory notebook, uploaded to a LIMS, or even displayed on the refrigerator for easy reference.
