The power of powder - Screening of protein-based pharmaceuticals

Proteins often form microcrystalline precipitates with an average size of crystallites (~ 0.1 - 1 μm) which in many cases are ideal for X-ray powder diffraction (XRPD). To date, a series of experiments and data analyses have been carried out, which demonstrate the validity of XRPD to study microcrystalline protein samples.

This method is powerful for high-throughput polymorph identification and crystal screening and also suitable for structure solution. In this webinar, we demonstrate the complementarity of laboratory and synchrotron XRPD as an analysis tool in protein crystallography. Parametric measurements (variation of relative humidity or pH) will be presented on pharmaceutical proteins and their complexes with organic ligands.

Irene Margiolaki is assistant professor at the Department of Biology of the University of Patras (UPAT, Patras, Greece). She completed a D.Phil in 2004 at the University of Sussex, UK. Between 2003 and 2010, she was a post-doc and later instrument scientist at the European Synchrotron Radiation Facility (ESRF), in Grenoble, France. Since 2010, she leads her 15 membered research team. Her work to date has been awarded by internationally distinguished organizations:

  • 2003: “Blue John Crystal” award from the biological structures of the British Crystallographic Association (BCA)
  • 2006: European Powder Diffraction Conference (EPDIC 10) young scientist award
  • 2010: UNESCO-L’Oréal co-sponsored international fellowship for Young Women in Life Sciences

Dr. Detlef Beckers did his PhD in Physics in 1996 at the Solid State Physics Institute in the Research Center Jülich in Germany. He joined PANalytical B.V. in 1996 (at that time Philips Analytical) in the position of Project Manager XRD (X-ray diffraction). In 2003 he became Market Segment Manager Pharmaceuticals, Food and Life Science with the responsibility for business and applications development in this segment. In 2009 he was additionally appointed as Product Manager and responsible for the market introduction of the Empyrean system. His current position is Lead Scientist XRD within PANalytical.
