NEB announces introduction of one-step cloning and multiple DNA fragment assembly

New England Biolabs (NEB®) announces the introduction of the NEBuilder HiFi DNA Assembly Cloning Kit and Master Mix for one-step cloning and multiple DNA fragment assembly in as little as 15 minutes. The novel, proprietary DNA polymerase in the NEBuilder HiFi enzyme mix results in extremely accurate fragment assemblies and cloning junctions, and can assemble DNA fragments ranging from 100 bp to at least 19 kb. The unique enzyme formulation of NEBuilder HiFi, which has numerous benefits over the Gibson Assembly® Master Mix (also available from NEB), further allows the joining of DNA fragments by use of a synthetic oligonucleotide splint. Experimental data highlighting the superior performance and quality of NEBuilder HiFi is available at NEBuilderHiFi.com. In addition, no licensing fees from NEB are required for commercial organizations using NEBuilder HiFi.

"NEB has supplied DNA assembly reagents for over 40 years and the new NEBuilder HiFi DNA Assembly Master Mix is the result of our commitment to producing high quality, innovative cloning reagents. The same reagent allows fast, virtually error-free standard cloning, as well as large, complex multi-fragment DNA assemblies. We continue to find new applications in which it excels and saves us valuable research time," says Tom Evans, Ph.D., who leads the DNA Enzymes Division at NEB.

NEB also provides a free tool, called the NEBuilder® Assembly Tool (nebuilder.neb.com), to simplify the design of primers. Scientists using fragments designed using other seamless cloning methods can easily assemble these with NEBuilder HiFi, due to compatibility of the fragments.
