The NKG2 (CD159) family of proteins includes NKG2D and NKG2A. The NKG2 proteins are significantly expressed on cytotoxic immune cells and form new appealing targets for developing immune checkpoint modulators.
The NKG2A is known as an inhibitor receptor and builds a heterodimer with CD94 (KLRD1). CD94 or NKG2A mixture reacts with the non-classical MHC class I molecule HLA-E on target cells.
This interaction prevents the triggering of T cells and NK cells through the tyrosine phosphates SHP-1 or SHP-2 signaling cascades.
Cancer cells use CD94/NKG2A-HLA-E, similar to how they use other immune checkpoints like PD-1/PD-L1, to evade the immune system.
NKG2D acts as an activating receptor. The activation of NKG2D by its ligands — such as MICA, MICB and ULBPs (ULBP1, ULBP2, ULBP3, ULBP4, ULBP5, ULBP6) — in human and H60 (a–c), RAE(α–ε) and MULT1 families in mice is a key antitumor signaling axis.
Using adaptor molecules, such as DAP10 and DAP12, the NKG2D protein transmits its signal transduction. By contrast, in humans, the NKG2D interacts mainly with DAP10.
In mice, NKG2D is also capable of controlling cytokine secretion (for example, IL-10, IL-9, IL-13). It also performs a crucial role in the fight against virus infection.
