VEGFs and their receptors: Key mediators in the nervous and vascular systems

Vascular endothelial growth factors, or VEGFs, constitute a class of secreted polypeptides that exhibit angiogenic and mitogenic activities. Due to the significance of VEGFs and receptors in tumorigenesis, angiogenesis and neuroprotection, they are attractive targets for drug development.

The members of the VEGF family — VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, VEGF-F and PlGF — bind in particular patterns to three associated receptor protein tyrosine kinases, namely, VEGFR1/FLT1, VEGFR2/KDR and VEGFR3/FLT4.

They also bind in specific patterns with neuropilin-1 (NRP1) and neuropilin-2 (NRP2) — two non-tyrosine kinase transmembrane proteins.

The ligand-receptor binding stimulates the formation of homomeric and heteromeric receptor complexes and triggers downstream signaling.

Apart from VEGF signaling, neuropilins also serve as co-receptors to various other signaling pathways, such as those induced by Hepatocyte Growth Factor (HGF), Epidermal Growth Factor (EGF), Platelet-Derived Growth Factor (PDGF), Fibroblast Growth Factor (FGF), Transforming Growth Factor beta (TGFβ) and Insulin-like Growth Factor (IGF).

VEGF family ligands and receptors
