Pharmaceutical density measurements are governed by a set of global pharmacopeial standards that ensure consistency, precision, and patient safety. This article is an overview of essential international and regional standards, outlining the permitted methods and measurement settings that laboratories must follow to remain compliant with quality control workflows.
Many aspects of global pharmacopeias have become standardized as global production systems have matured. However, based on their location, pharmaceutical manufacturers may be obliged to conform with one or more of the following requirements when using density meters for quality control activities.
European Pharmacopoeia (Ph. Eur.)
Legally binding in 39 European countries and utilized in more than 120 countries worldwide.
Ph. Eur. 2.2.5 - Relative density can be determined using a density bottle (solids or liquids), a hydrometer (liquids), a hydrostatic balance (solids), or a digital density meter with an oscillating transducer (liquids and gases).
International Pharmacopoeia (Ph. Int.)
Ph. Int. 1.3 - At 20 °C, use a hydrostatic balance or pycnometer to determine mass density, relative density, and weight per milliliter.
Pharmacopoeia of the People’s Republic of China (Chinese Pharmacopoeia)
This includes both traditional Chinese and Western medicine.
Chinese Pharmacopeia 0601 - To measure relative density, use a specific gravity bottle at 20 °C (method I), a hydrostatic balance at 20 °C (method II), or a digital density meter with an oscillating transducer (method III).
United States Pharmacopoeia (USP)
USP Chapter 841 (as of USP 29), Specific Gravity Method II - Measure specific gravity at 25 °C using a pycnometer or an oscillating transducer density meter (digital density meter).
The primary goal of these standards is to enable high-quality, consistent measurements that improve consumer safety when utilizing pharmaceutical products.
