Lengthy treatment with low-dose 6-mercaptopurine inhibits resistant triple-negative breast cancer cells

The cover for issue 7 of Oncotarget features Figure 5, "SUM149-MA cells surviving a 6-MP treatment are sensitive to chemotherapeutic drugs," published in "Inhibition of resistant triple-negative breast cancer cells with low-dose 6-mercaptopurine and 5-azacitidine" by Singh, et al. which reported that the authors have reported that a lengthy treatment with low-dose 6-mercaptopurine, a clinically useful anti-inflammatory drug, inhibits such resistant cells.

The World Health Organization (WHO) has labeled the double mutant B.1.617 variant — first discovered in India —a variant of concern for its high transmission rate and ability to evade immune responses. New research posted to the preprint server bioRxiv finds B.1.617 can modestly reduce neutralizing antibody responses generated from the Pfizer-BioNTech vaccine.

Based on structural analysis, researchers found that vitamin C binds to 3CLpro, the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) main protease essential in viral replication. A team of researchers from the University of Virginia, USA, used single-virus fusion experiments with exogenously controlled proteases to investigate the mechanism of activation directly. They found that the subcellular location of entry depends on the protease activity of the various components, and parallel pathways are possible.
