Transgenic mouse model validates mutant IDH2 protein as candidate for targeted anticancer therapies

In recent years, mutations in two metabolic enzymes, isocitrate dehydrogenase-1 and 2 (IDH1 and IDH2), have been identified in approximately 20 percent of all acute myeloid leukemias (AML). As a result, mutant IDH proteins have been proposed as attractive drug targets for this common form of adult leukemia.

Kolltan Pharmaceuticals, a privately held biopharmaceutical company, today announced treatment of the first cancer patient with Kolltan's lead drug candidate, KTN3379, as part of a Phase 1 clinical study evaluating safety, preliminary evidence of antitumor activity, and pharmacokinetics. KTN3379 is a human monoclonal antibody that blocks the activity of ErbB3, a member of the ErbB family of receptor tyrosine kinases.

ARIAD Pharmaceuticals, Inc. today announced the commercial availability of Iclusig (ponatinib) for adult patients with refractory chronic myeloid leukemia and Philadelphia-chromosome positive acute lymphoblastic leukemia in the United States. ARIAD has begun shipping Iclusig to Biologics, Inc., its exclusive specialty pharmacy, which is now filling prescriptions from physicians and distributing the cancer medicine to patients.

Research opens door to new direction of PAH treatment

The development of new, more effective vasodilators to treat pulmonary arterial hypertension has been hampered because of their systemic toxicity and adverse side effects. An international team of investigators seeking to surmount these problems and increase drug efficacy have determined that a vascular homing peptide can selectively target hypertensive pulmonary arteries to boost the pulmonary but not systemic effects of vasodilators. Importantly for potential clinical use, this peptide retains its activity when given sublingually. The results using a rat model of PAH are published in the American Journal of Pathology.

Horizon, AstraZeneca partner to explore oncology-relevant genotypes to identify novel drug targets

Horizon Discovery and AstraZeneca have entered into a research, collaboration and license agreement to explore a range of oncology-relevant genotypes with the aim of identifying and validating a number of novel drug targets. This deal is the second collaboration between AstraZeneca and Horizon, and follows the announcement in April 2013 of an oncology discovery program to explore Horizon's first-in-class kinase target program, HD-001.

Mannotriose regulates learning and memory signal transduction in hippocampus

The main component of the Chinese herb Rehmannia, mannotriose, can improve learning and memory. Dr. Lina Zhang and colleagues from Shanghai University of Traditional Chinese Medicine in China used 1 - 10 mol/L mannotriose to treat primary hippocampal neurons insulted with 1 - 10 mol/L corticosterone.
