Positive results from PLX4032 Phase 3 Trial in patients with metastatic melanoma

Plexxikon today announced positive data from an interim analysis of the BRIM3 trial, a large multi-center Phase 3 clinical study of PLX4032 in patients with previously untreated metastatic melanoma with the BRAF mutation. Patients treated with PLX4032 had an improved overall survival compared to patients treated with dacarbazine, the current standard of care.

A new investigational drug designed to stop cancer cells from reproducing may offer hope for patients with advanced solid tumor cancers. Clinical trials of TKM-PLK1 for qualified patients are now open at the Virginia G. Piper Cancer Center at Scottsdale Healthcare.

Pfizer initiates rolling submission of crizotinib NDA for ALK-positive advanced NSCLC

Pfizer Inc. announced today that it has initiated the rolling submission of a New Drug Application (NDA) to the U.S. Food and Drug Administration (FDA) for crizotinib (PF-02341066), an oral first-in-class anaplastic lymphoma kinase (ALK) inhibitor for the treatment of patients with advanced non-small cell lung cancer (NSCLC) whose tumors are ALK-positive. Pfizer expects to complete the submission in the first half of 2011.

Daiichi Sankyo and ArQule initiate enrollment in ARQ 197 Phase 3 trial in NSCLC

Daiichi Sankyo Company, Limited and ArQule, Inc. today announced that the first patient has been enrolled in the Phase 3 trial of ARQ 197, an investigational selective inhibitor of the c-Met receptor tyrosine kinase, in combination with erlotinib, for patients diagnosed with non-squamous, non-small cell lung cancer (NSCLC) who have received one or two prior systemic anti-cancer therapies.

Plexxikon, Genentech enter PLX4032 co-promotion agreement

Plexxikon, Inc., announced today that it has signed an agreement with Genentech to co-promote PLX4032 (RG7204) in the United States. Blocking activation of delta-PKC enzyme could protect lungs from neutrophil-mediated damage

All of us may be able to breathe a little easier now that scientists from Pennsylvania have found a new therapeutic target for controlling dangerous inflammation in the lungs. A new research report in the January 2011 issue of the Journal of Leukocyte Biology (http://www.jleukbio.org) suggests that blocking the activation of an enzyme called delta-protein kinase C (delta-PKC) could protect the lungs from neutrophil-mediated damage, which can result in out of control inflammation.

Sequestering GSK3 enzyme leads to stabilization of proteins in cells

The cell signaling pathway known as Wnt, commonly activated in cancers, causes internal membranes within a healthy cell to imprison an enzyme that is vital in degrading proteins, preventing the enzyme from doing its job and affecting the stability of many proteins within the cell, researchers at UCLA's Jonsson Comprehensive Cancer Center have found.

Researchers discover new way to prevent spread of cancer

Scientists now know that some cancer cells spread, or metastasize, throughout the body the old-fashioned way -- by using their feet. But researchers at Duke Cancer Institute have discovered a way to short-circuit their travels by preventing the development of these feet, called invadopodia. This discovery is even more important because preventing the development of these "feet" also eliminates the action of proteins present in the feet that burn through intact tissue and let cancer cells enter new cells.
