Generative AI-driven drug Rentosertib enters Phase III trial for idiopathic pulmonary fibrosis

Insilico Medicine, a clinical-stage biotechnology company powered by generative AI, today announced that it has dosed the first patient with Rentosertib (known as ISM001-055 / INS018_055) in GENESIS-IPF-3, a Phase III clinical trial (NCT07687459, CTR20262475) at Peking Union Medical College Hospital, while Shanghai Pulmonary Hospital has also enrolled its first patient on the same day.

Tizanidine looked promising early in a migraine trial, but the benefit did not last

A randomized, double-blind, placebo-controlled trial found that low-dose tizanidine did not provide sustained reductions in headache burden over 12 weeks in people with episodic migraine, despite a transient improvement with the 1 mg dose at week 4. Exploratory analyses suggested that smoking and sex may influence tizanidine exposure, while sex-specific efficacy signals remained hypothesis-generating and require confirmation.

Rentosertib shifts multiple proteomic aging clocks toward younger age profiles in IPF trial

Researchers applied six proteomic aging clocks to serum samples from a phase 2a trial of rentosertib in people with idiopathic pulmonary fibrosis and found treatment-associated shifts toward younger predicted biological-age profiles, particularly with the 30 mg twice-daily regimen. Additional proteomic analyses linked treatment to changes in senescence, metabolism, growth-factor signaling, and fibrosis-related pathways, although the study could not determine whether these effects reflected genuine geroprotection rather than disease modification.

Psilocybin microdosing sounds promising, but the strongest studies tell a different story

A systematic review of 11 studies involving 3,262 adults found that psilocybin mushroom and truffle microdosing was associated with reported improvements across areas including mental health, well-being, cognition, creativity, and social functioning. However, the evidence was methodologically weak and highly heterogeneous, the two placebo-controlled trials found no significant improvements, and inadequate adverse-event reporting means efficacy and safety remain uncertain.

Semaglutide did more than cut weight in this analysis of 6,239 adults

A systematic review and meta-analysis of nine randomized controlled trials involving 6,239 adults with overweight or obesity without diabetes found that subcutaneous semaglutide produced substantially greater reductions in body weight, waist circumference, and C-reactive protein than placebo. The higher dose of 7.2 mg semaglutide showed potential for greater weight loss, although the evidence is largely based on a single trial and requires confirmation.
