Study finds new therapeutic target for polycystic kidney disease

In work suggesting a new approach to treating polycystic kidney disease (PKD), a leading cause of kidney failure, researchers at Children's Hospital Boston were able to block the formation of fluid-filled cysts, the hallmark of the disease, in a mouse model. Their findings, using a compound that inhibits a receptor known as c-Met, will be published in the September 13th online edition of the Journal of Clinical Investigation.

New gene required for memory formation identified

A team led by a Scripps Research Institute scientist has for the first time identified a new gene that is required for memory formation in Drosophila, the common fruit fly. The gene may have similar functions in humans, shedding light on neurological disorders such as Alzheimer's disease or human learning disabilities.

PLX4032 trial shows high degree of selectivity to achieve tumor shrinkage

Plexxikon today announced publication of key data in this week's edition of Nature demonstrating that the high degree of selectivity of PLX4032 (RG7204) enables substantial RAF/MEK/ERK pathway inhibition, which may be necessary to achieve significant tumor shrinkage and clinical response in patients with BRAF mutant melanoma.

Quick increase of calcium concentration in cells rapidly trigger Aurora-A kinase enzyme activity

Aurora-A kinase (AurA) is an enzyme that is hyperactive in many cancers and drives tumor cell proliferation. Several AurA inhibitors are currently being tested in clinical trials to see if they slow tumor growth. Now, researchers in the Developmental Therapeutics Program at Fox Chase Cancer Center have identified an activation signal for AurA. They report in the September 7 issue of Nature Communications that a quick increase in the calcium concentration in a cell rapidly triggers AurA kinase activity.

Researchers develop new RNA molecule approach to kill cancer cells

Researchers at the California Institute of Technology (Caltech) have engineered a fundamentally new approach to killing cancer cells. The process developed by Niles Pierce, associate professor of applied and computational mathematics and bioengineering at Caltech, and his colleagues uses small RNA molecules that can be programmed to attack only specific cancer cells then, by changing shape, those molecules cause the cancer cells to self destruct.

Gene mutations that express LRRK2 linked to increased risk of Parkinson's disease: Researchers

A significant number of Parkinson's disease patients have a mutation of the enzyme Leucine-Rich Repeat Protein Kinase 2 (LRRK2, also known as dardarin). However, little is understood about how it is regulated or functions. In a new paper in the Signal Knowledge Environment of the Biochemical Journal, Dario Alessi and colleagues from the University of Dundee demonstrate that a family of proteins, the 14-3-3 proteins, interact with LRRK2.

Research on OGF-OGFr complex helps to understand pathobiology of diseases

Researchers at The Pennsylvania State University College of Medicine, Hershey, Pennsylvania have discovered that the Opioid Growth Factor (OGF, [Met5]-enkephalin) and its receptor, OGFr, a clinically important system with potent antitumor properties, has controlled entry from the cytoplasm to the nucleus. The nucleocytoplasmic passage of OGF-OGFr is critical to cell proliferation and suggests that there are hierarchical levels of nuclear import.

Researchers find new drug to treat circadian rhythm disorders

Researchers funded by the Biotechnology and Biological Sciences Research Council (BBSRC) and the Medical Research Council (MRC) have successfully used a drug to reset and restart the natural 24 hour body clock of mice in the lab. The ability to do this in a mammal opens up the possibility of dealing with a range of human difficulties including some psychiatric disorders, jet lag and the health impacts of shift work.

Study supports role for SNARK protein in regulating glucose transport during muscle contraction and exercise

Skeletal muscles combust both lipids and carbohydrates during exercise. The carbohydrates consist of both glycogen stored in the muscles as well as glucose extracted from the blood. Being a major sink for glucose disposal, skeletal muscle represents an important model tissue for studying the intracellular signaling pathways leading to increased glucose transport.
