Effective bioanalysis of these molecules necessitates rigorous technique development, optimized sample preparation, thorough stability evaluation, and advanced mass spectrometry expertise to provide precise, repeatable pharmacokinetic and toxicokinetic data across preclinical and clinical studies.

Semaglutide poses multiple bioanalytical challenges due to its peptide structure (4.1 kDa), extensive albumin binding, and tendency for non-specific adsorption. Achieving robust quantification requires precise optimization of sample preparation to maximize recovery while minimizing matrix impacts and analyte loss.
Additional challenges involve selecting appropriate precursor and product ions from several charge states, maintaining analyte stability during sample handling, and achieving the sensitivity needed to support pharmacokinetic research across a broad concentration range. This frequently requires a custom LC-MS/MS strategy tailored to the distinct physicochemical characteristics of the molecule.
