Interleukin 6 (IL-6) is a pro-inflammatory cytokine involved in bone metabolism, hematopoiesis, immune responses and embryonic development. High IL-6 is noted in patients with microbial infections and other inflammatory conditions.
Targeting IL-6 has significant clinical implications for managing, diagnosing and preventing infectious diseases.
The pro-inflammatory function of IL-6 has been controlled by a trans-signaling process involving soluble IL-6R (sIL-6R) and gp130 receptor. The production of sIL-6R is triggered by TNF-alpha and IL-1beta. These two pro-inflammatory cytokines provoke the proteolytic cleavage of IL-6R by triggering ADAM10 and ADAM17.
Dissimilar to the majority of the other soluble receptors (for example, IL-1RA and IL-18BP), sIL-6R is NOT an inhibitor of IL-6. Rather, it triggers IL-6 signaling by developing a complex with IL-6 and gp130.

IL-6 illustrates restricted cross-species bioactivity. Human IL-6 has the ability to attach to both human and mouse IL-6R, but mouse IL-6 can only fix to mouse IL-6R.
