IL-6: A Critical Inflammatory, Pleiotropic Cytokine

IL-6 is a crucial pleiotropic inflammatory cytokine. Increased IL-6 is seen in patients who are suffering from microbial infections and other similar inflammatory conditions. The serum level of IL-6 is usually associated with the severity of the inflammation.

IL-6’s pro-inflammatory function is controlled by a trans-signaling process that involves the gp130 receptor and soluble IL-6R (sIL-6R). TNF-alpha and IL-1beta trigger the production of sIL6R. Both these pro-inflammatory cytokines promote the proteolytic cleavage of IL-6R by stimulating ADAM17 and ADAM10.

sIL-6R is different from most other soluble receptors (for example, IL-18BP and IL-1RA) in that it does not inhibit IL-6. Rather, it forms a complex with gp130 and IL-6 to trigger IL-6 signaling.

Besides this, IL-6 can transduce its signal via membrane-bound IL-6R. For this kind of signal transduction, gp130 also needs to be the co-receptor.

Recombinant membrane-bound IL-6 receptor: IL-6R

IL-6 shows restricted cross-species bioactivity. Human IL-6 can attach to both human and mouse IL-6R; however, the mouse IL-6 can attach only to mouse IL-6R.

Human IL-6R neutralizing monoclonal antibody neutralizes IL-6-dependent inhibition of cell proliferation. The IC50 is generally 0.15–0.6 μg/mL.

Interleukin 6 (IL6, also called IFNB2, IFN-beta-2, BSF2, BSF-2, IL-6, HSF, HGF and CDF) is found on the choromosome 7p15.3. Interleukin 6 is conserved in Rhesus monkeys, chimpanzees, chickens, rats, mice and dogs.

The gene creates a 23718 Da protein made up of a total of 212 amino acids. IL-6 background

As a multifunctional α-helical cytokine, Interleukin-6 (IL-6) controls the growth and differentiation of cells in numerous tissues. It is specifically known for its role in acute phase reactions and the immune response.

The IL-6 protein is produced by different types of cells, such as macrophages and T cells, as a phosphorylated and unevenly glycosylated molecule. It applies actions through its heterodimeric receptor made up of IL-6R that does not contain the tyrosine/kinase domain and attaches IL-6 with low affinity.

The glycoprotein 130 (gp130) is universally expressed and binds the IL-6. IL-6R complex has an excellent affinity and hence transduces signals.

IL-6 plays a key role in bone metabolism, hematopoiesis and cancer development. It has also been defined as a crucial factor in guiding the change from innate to acquired immunity.
